Understanding the genetic mechanisms for cadmium (Cd) uptake and translocation in common wheat (Triticum aestivum) is of significance in food Cd contamination control. In this study, a diverse panel of 132 wheat cultivars was collected from the North China Plain. The cultivars were evaluated in terms of their phenotypic variations in response to Cd stress and subjected to a genome-wide association study (GWAS) to identify single nucleotide polymorphisms (SNPs) associated with the phenotypic variations at the seedling stage. Significant phenotypic variations with high heritability were observed among the wheat cultivars exposed to 40 μmol L-1 Cd for the studied traits, including root length (RL), shoot length (SL), root and shoot dry biomasses (RDW and SDW, respectively), root and shoot Cd concentrations (RCD and SCD, respectively), and Cd translocation factor (TF). Mean RCD, SCD, and TF ranged from 1.0 to 33.8, 0.125 to 2.022, and 0.009 to 0.321 mg g-1, respectively. Cluster analysis showed that wheat cultivars with higher RL, SL, RDW, and SDW under Cd stress were able to accumulate more Cd in root, leading to a lower Cd TF. Mixed linear model-based association analysis detected 17 novel significant marker-trait associations (MTAs), four of which were significant at a genome-wide scale. Most of the significant MTAs controlled Cd TF and explained 17.17%-26.47% of the phenotypic variations. Some of the SNP loci were physically close to a reported Cd-related quantitative trait locus or gene on wheat chromosomes. Results of this study provided a list of wheat cultivars with the potential of low Cd accumulation and enriched our knowledge on the genetic basis of Cd uptake and translocation in wheat. Pyramiding breeding of superior alleles detected in this study may additionally reduce Cd accumulation of improved wheat cultivars with excellent agronomic traits.  相似文献

This study is about relationships between total extractive contents, heartwood colour and light-induced discolourations of the tropical species African Padauk (Pterocarpus soyauxii Taub) and Jatoba (Hymenaea courbaril L.). Wood surface colour was measured spectrophotometrically before and after light irradiation. Subsequently, the total extractive content was determined and linked to the colour parameters. Total extractive content varied between 6 % (Jatoba) and 27 % (African Padauk). The fact that non-irradiated Jatoba appeared lighter and more yellow with higher extractives contents, opposed the often assumed relationship that more heartwood extractives cause darker wood. For light-irradiated Jatoba wood only the reddishness was more intense when samples had higher extractive content. In contrast, light-irradiated African Padauk showed significant differences in all colour parameters as extractives varied. High overall colour change in African Padauk due to light irradiation was observed; however, no relationship with the extractive content existed. In contrast, discolouration rate of Jatoba wood strongly varied with its extractive content. Individual wood species react differently during exposure to light and therefore require specific surface treatment to maintain the desired colour appearance and colour stability.  相似文献

The aim of this study was to assess fungal communities in roots of healthy-looking Pinus sylvestris L. and Picea abies (L.) Karst. seedlings in nine forest nurseries in Sweden using a combination of traditional culturing and direct sequencing of internal transcribed spacer of fungal ribosomal RNA (ITS rRNA) from the roots. Culturing from 1800 surface-sterilised root segments resulted in 2387 fungal cultures representing 42 different taxa. Direct sequencing from 180 root segments resulted in 119 ITS rRNA sequences representing 25 different taxa. In total, 55 different fungal taxa were detected using both methods. Although direct sequencing was more efficient than culturing in detecting different fungal taxa, both methods provided complementary information about fungal communities in roots since each detected rather different groups of fungi. The most dominant taxa detected by culturing were Trichoderma viride Pers. (19.5%), Phoma mucivora Davey & Currah (19.1%), Phialocephala fortinii Wang & Wilcox (17.4%) and Meliniomyces variabilis Hambl. & Sigler (10.2%), while Thelephora terrestris Ehrh. (26.1%), Unidentified sp. NS126 (25.2%) and Heliotales sp. C20 (10.1%) were most commonly detected by direct sequencing. In conclusion, results showed that forest nurseries in Sweden harbour diverse communities of fungi associated with the roots of healthy-looking P. sylvestris and P. abies seedlings. Although fungal communities were often dominated by saprotrophs and endophytes, several facultative pathogens were also detected indicating that under suitable conditions they may be a potential threat to the plants.  相似文献

  1. Marine protected areas (MPAs) are becoming a widely used tool for the conservation of biodiversity and for fishery management; however, most of these areas are designed without prior knowledge of the basic ecological aspects of the species that they are trying to protect.
  2. This study investigated the movement of two top predators: the Galapagos shark, Carcharhinus galapagensis, and the yellowtail amberjack, Seriola lalandi, in and around the Motu Motiro Hiva Marine Park (MMHMP) using MiniPAT satellite tags to determine the effectiveness of this MPA for the protection of these species.
  3. The Galapagos sharks (n = 4) spent most of their tag deployment periods inside the MMHMP. However, high intraspecific variability was observed in their movement dynamics. Daily individual maximum movements ranged from 17 to 58 km and the maximum distance from Salas y Gómez Island, the only emergent island within the MMHMP, ranged from 31 to 139 km.
  4. The maximum linear distance travelled for a female juvenile Galapagos shark (152 cm total length) was 236 km, which is greater than the maximum distance previously documented for juveniles of this species (<50 km).
  5. For the yellowtail amberjack (n = 1), 91% of the satellite geolocations were within the MMHMP, with a maximum daily distance travelled of 6 km. The maximum distance travelled between points was 111 km and the maximum distance from Salas y Gómez Island was 62 km.
  6. All archival tagged fish spent most of their time at depths of <50 m and never left the epipelagic zone. Daytime versus night-time differences were pronounced in all individuals but showed high interindividual variability.
  7. This study provides a baseline on the movement of these two top predators in the MMHMP and provides valuable insights for the creation of MPAs in the region and elsewhere.

OBJECTIVE: To assess the efficacy of prophylactic hoof health examination and trimming during midlactation at reducing the incidence of lameness during late lactation in dairy cows. DESIGN: Randomized field trial. ANIMALS: 333 Holstein cows. PROCEDURES: Cows without apparent lameness were randomly allocated into 1 of 2 groups approximately 204 days after calving. Cows allocated to the treatment group (n = 161) were examined on a tilt table for diagnosis and underwent hoof-trimming procedures, if needed, for treatment of hoof disorders or lesions. Cows in the control group (n = 172) were not examined. Cows were assigned a locomotion score weekly for 28 weeks after allocation to a group. The number of cows classified as lame during late lactation (approx 205 to 400 days after calving) was compared between groups to assess the efficacy of prophylactic examination and trimming. RESULTS: Incidence of lameness during late lactation was 24% in cows in the control group and 18% in cows in the treatment group. CONCLUSIONS AND CLINICAL RELEVANCE: The 25% decrease in number of new cases of lameness in cows undergoing prophylactic hoof health examination and trimming during midlactation may be relevant for the well-being of dairy cows and should not represent a major economic burden to producers.  相似文献

The Okavango catchment in southern Africa is subject to environmental as well as socio‐economic transformation processes such as population growth and climate change. The degradation of soil and vegetation by deforestation and overgrazing is one of the downsides of this development, reducing the capacity of the land to provide ecosystem functions and services. In this study, climate simulations are brought together with secondary socioeconomic, pedologic and remote‐sensing data in a GIS‐based assessment of the factors commonly associated with land degradation risk. A high resolution overview is provided for decision‐makers and stakeholders in the region by identifying priority intervention areas where a long‐term decline in ecosystem function and land productivity is most likely to occur. The approach combines 19 risk factors into seven individual ratings for topography, landcover, soil, demography, infrastructure, livestock pressure and climate. These ratings are then weighted and combined into an integrated degradation risk index (DRI). The results show that the land degradation risk is quite heterogeneously distributed in the study area and caused by different factors. Three hot‐spots are identified and compared, one of which is in the far northwestern part of the catchment, one around the local center Rundu and one on the outskirts of the Okavango Delta. We conclude that the approach is suitable to give an overview on degradation risk in the study area, although the classification process is a crucial procedure that should be standardized for further research. Campylobacter is the second leading cause of foodborne illness in the United States. Although many food production animals carry Campylobacter as commensal bacteria, consumption of poultry is the main source of human infection. Previous research suggests that the biology of Campylobacter results in complete flock colonization within days. However, a recent systematic review found that the on-farm prevalence of Campylobacter varies widely, with some flocks reporting low prevalence. We hypothesized that the low prevalence of Campylobacter in some flocks may be driven by a delayed introduction of the pathogen. The objectives of this study were to (a) develop a deterministic compartmental model that represents the biology of Campylobacter, (b) identify the parameter values that best represent the natural history of the pathogen in poultry flocks and (c) examine the possibility that a delayed introduction of the pathogen is sufficient to replicate the observed low prevalence examples documented in the literature. A deterministic compartmental model was developed to examine the dynamics of Campylobacter in chicken flocks over a 56-day time period prior to movement to the abattoir. The model outcome of interest was the final population prevalence of Campylobacter at day 56. The resulting model that incorporated a high transmission rate (β = 1.04) was able to reproduce the wide range of prevalence estimates observed in the literature when pathogen introduction time is varied. Overall, we established that the on-farm transmission rate of Campylobacter in chickens is likely high and can result in complete colonization of a flock when introduced early. However, delaying the time at which the pathogen enters the flock can reduce the prevalence observed at 56 days. These results highlight the importance of enforcing strict biosecurity measures to prevent or delay the introduction of the bacteria to a flock.  相似文献

Summary The tolerance of aluminum (Al) of disomic substitution lines having the chromosomes of the D genome of Triticum aestivum L. cv. Chinese Spring individually substituted for their homoeologues in T. turgidum L. cv. Langdon was investigated by the hematoxylin method. The disomic substitution lines involving chromosome 4D were more Al tolerant than Langdon. The tolerance was found to be controlled by a single dominant gene, designated Alt2, that is in the proximal region of the long arm of chromosome 4D. The locus was mapped relative to molecular markers utilizing a population of recombinant chromosomes from homoeologous recombination between Chinese Spring chromosome 4D and T. turgidum chromosome 4B. Comparison of the location of Alt2 in this map with a consensus map of chromosomes 4B and 4D based on homologous recombination indicated that Alt2 is in a vicinity of a 4 cM interval delineated by markers Xpsr914 and Xpsr1051. The Alt2 locus is distal to marker Xpsr39 and proximal to XksuC2. The Altw locus is also proximal to the Knal locus on chromosome 4D that controls K+/Na+ selectivity and salt tolerance. In two lines, Alt 2 and Knal were transferred on a single 4D segment into the long arm of T. turgidum chromosome 4B.  相似文献
